Estimating colour differences for different areas of image

I’m working with the very similar colour images to those which you can get from the photos of the “Eyes” app in the iOS app store.
To explain what I’m trying to achieve, I am trying to analyse the “Eyes” app and estimate the colour differences between pairs of regions of the image, so that I can find the colour areas of the photo with similar colours to the “Eyes” app.
I have managed to extract the colour difference between pairs of regions, but I can’t get the colour differences to appear on the image – they simply show up as black boxes.
I’m using python to make the analysis.
